Optimization of methods of DNA extraction from wheat genotypes

Solation of the DNA of many plant species is considered difficult due to the high concentration of secondary metabolites, such as polysaccharides and polyphenols. There are several methods to solve this problem. In our research, we conducted an analysis of existing methods for DNA extraction from plants. Disruption of cells is usually carried out by mechanical means in the presence of aqueous solutions of detergents dissolving cellular membranes, and chelating agents that suppress the activity of nuclease. Methods of DNA extraction for molecular genetic studies. As a result, selected was a potassium- acetate method , which allowed to obtain not only highly pure, but also the highly concentrated preparations of DNA. The frequency of DNA ranged 1.95 to 2.05. DNA concentration ranged from 361.5 to 3571.8 ng/mkl depending on the genotype of wheat.References
