Создание рекомбинантных штаммов дрожжей для получения биоэтанола из целлюлозосодержащего сырья

Аннотация

С помощью генно-инженерных методов сконструирован pHO-GAPDH-eng1-GAPDH-α-cel7A-myc-6xHis-GAPDH-α-bglI-flag-KanMX4-HO, содержащий гены эндо-1,4-β-глюканазы, целлобиогидролазы и β-гликозидазы. Каждый ген в составе интегрального вектора содержал  сигнальный пептид α-фактора дрожжей и экспрессируются под контролем промотора глицеральдегид-3-фосфат-дегидрогеназы (GAPDH). Конструированный вектор обеспечивает интеграцию генов целлюлитических ферментов в хромосому дрожжей путем генного замещения с использованием участка гомологичной последовательности, НО гена.

Созданы новые стабильные штаммы дрожжей с генами целлюлитических ферментов в HO локусе хромосомы дрожжей. Хромосомная интеграция генов эндо-1,4-β-глюканазы, целлобиогидролазы и β-гликозидазы в НО локус генома дрожжей была подтверждена методом ПЦР. Данный штамм обеспечивает непрерывную экспрессию генов целлюлитических ферментов и секрецию продуктов  экспрессии в окружающую среду. Показано, что рекомбинантные штаммы S.cerevisiae, экспрессирующие гены целлюлаз способны расти в синтетической среде, содержащей целлобиозу или карбоксиметилцеллюлозу (CMC) в качестве единственного источника углерода. Кроме того, рекомбинантный штамм производит 15,6 г/л этанола из 20% целлобиозы и 20% CMC. Эти результаты свидетельствуют о том, что рекомбинантные штаммы S.cerevisiae, могут быть применимы к одновременному осахариванию и ферментации целлюлозосодержащей биомассы.
