Запасные белки зерновки как маркеры в селекции первого казахстанского сорта риса с окрашенным перикарпом

ӨББИ коллекциясының 11 сорттарына, 9 коллекциялық үлгілеріне және 12 гибридтеріне белоктық паспортизация жүргізілді. Қор белоктарының электрофорезі өзгертілген Laemmli әдісі бойынша жүргізілді. Күріш оризениндерін бөліп алу және фракциялау жағдайлары оңтайландырылды. Зерттеу жұмыстарының нәтижесінде қызыл дәнді Ко 296 R үлгісінде басқа қызыл дәнді формаларда айқын байқалмаған молекулалық массасы шамамен 55 kDа болатын қарқынды белок спектрі анықталды. Перикарпы боялған Қара күріш генотипінің қор белоктары спектрінде молекулалық салмағы 55 және 60 kDа тең белоктық компоненттер айқындалды, бұл белоктық компоненттер генотиптің басқа формалардан ерекшелігін көрсетеді. ӨББИ селекциясындағы F1 гибридтерінің «шынайылығы» ата-аналық формалардың компоненттерінің бар болуымен дәлелденді. Проведена белковая паспортизация 11 сортов, 9 коллекционных образцов и 12 гибридов риса из коллекции ИББР. Электрофорез запасных белков проводили модифицированным методом Laemmli. Подобраны оптимальные условия выделения и фракционирования оризенинов риса. В результате проведенных работ у краснозерного образца Ко 296 R выявлены интенсивные белковые спектры с молекулярной массой около 55 kDа, тогда как у других краснозерных форм данный компонент слабо выражено. В спектре запасных белков семян у генотипа Черный рис (Китай) с антоциановой окраской наблюдается проявление интенсивных белковых полос с молекулярной массой 55 и 60 kDа, что подтверждает сортспецифичность данного генотипа. Подтверждена «истинность» гибридов F1 поколений из 12 комбинаций селекции ИББР наличием компонентов обеих родительских форм. Seed storage protein identification of 11 rice varieties, 9 collection samples and 12 hybrids from the IPBB collection were conducted. Electrophoresis of storage proteins was carried out by method of Laemmli. Optimum conditions were selected for isolation and fractionation of rice glutelin. As a result of this research in red grain sample Ko 296 R revealed intense protein spectra with molecular weight of about 55 kDa, whereas the other red grain forms of this component weakly expressed. In the spectrum of seed storage proteins in genotype Black rice (China) with anthocyanin coloration observed manifestation of intense protein bands with molecular weights of 55 and 60 kDa, which confirms specificity of this genotype.The "verity" of F1 hybrids of 12 combinations from the IPBB breeding was confirmed by the presence of components of both parental forms.Библиографиялық сілтемелер
