Идентификация генетических маркеров лекарственной устойчивости на основе полных геномов клинических изолятов Mycobacterium tuberculosis

Проведено полногеномное секвенирование 20 клинических изолятов M. tuberculosis с использованием современных методов и инструментов секвенирования нового поколения. Большинство изученных изолятов M. tuberculosis (18) относятся к семейству Beijing (происхождение восточно-азиатское East Asian), только два изолята показали принадлежность к семействам Т (происхождение евро-американское Euro-American) и MANU-1 (индо-океанское Indo-Oceanic). Изучены генетические маркеры для диагностики лекарственно-устойчивых форм туберкулеза (MDR и XDR штаммы M. tuberculosis). Проведен анализ генетических локусов клинических изолятов M. tuberculosis определяющих устойчивость к противотуберкулезным лекарственным препаратам из числа найденных геномных вариантов, а также соответствующие гены. Обнаружено от 13 до 30 генетических локусов с геномными вариантами среди полногеномных данных 20 изолятов M. tuberculosis. Найдены геномные варианты в четырех генах PE_PGRS24, PPE24, PPE5, PE_PGRS56, кодирующие белки семейства РЕ/РРЕ и являющихся уникальными и представленными только для вида Mycobacterium, которые характерны только для MDR и XDR клинических изолятов. Белки данного семейства могут играть роль факторов вирулентности и способствовать успешному инфицированию.
