RAPD-fingerprinting of soil micromycetes

In this study stable RAPD-profiles for 7 soil micromycetes isolates were obtained by amplification of genomic DNA with single nonspecific primers. The arbitrary primer (decamer AP3), primer derived from the intergenic spacer region (T3B) and microsatellite primer (GTG)5 were applied in PCR. Precise and reproducible profiles of the amplification products were obtained for all seven micromycetes isolates. Species specificity of the amplicons number and size was demonstrated. In some cases, intraspecific variability was observed. The results show justification for the use of DNA fingerprinting for rapid identification and labeling of soil micromycetes, as well as evaluation of their stability during long-term cultivation.Downloads
