The cDNA gene, coding sheeppox virus (SPPV) (“NISCHI” strain) external envelope protein EEV122 has been cloned. Recombinant DNA-constructions «Y-EEV119-TMV» and «ARCх3-EEV119-ТМV have been constructed. The first recombinant construction «Y-EEV119-TMV» in its 5’-untranslated region (5’-UTR) contained 5’-UTR of Potato Virus Y (PVY) genomic RNA, and the second one «ARCх3-EEV119-ТМV» in its 5’-UTR contained three copy of 10- nucleotide sequence “ARC” (“ARCx3”), which was complementary to the central region of wheat 18S rRNA (nucleotides 1113-1122). Correspondent mRNAs have been synthesized in vitro, and these mRNAs were translated in wheat germ cell free system. It was shown, that both RNA-constructions provided a high level of SPPV EEV119 protein synthesis, though 5’-UTR of PVY gRNA provided higher translation level, than “ARCx3” sequence.Downloads
